Bisphosphonates impair the onset of bone formation at remodeling sites

چکیده
Bisphosphonates are widely used anti-osteoporotic drugs targeting osteoclasts. They strongly inhibit bone resorption, but also reduce formation. This reduced formation is commonly ascribed to the mechanism maintaining resorption/formation balance during remodeling. The present study provides evidence for an additional where bisphosphonates actually impair onset of after resorption. based on morphometric parameters recently developed assess activities reversing resorption Herein, we compare these in cancellous alendronate- and placebo-treated postmenopausal osteoporotic patients. Alendronate increases prevalence eroded surfaces characterized by reversal cells/osteoprogenitors at low cell density remote from active surfaces. indicates deficient expansion – event that indispensable start Furthermore, alendronate decreases coverage remodeling compartment canopies, a putative source cells/osteoprogenitors. Finally, activation frequency formation, more formative compared All correlate with each other. These observations lead model hamper osteoprogenitor recruitment required initiate effect results larger surface, thereby explaining well-known paradox without decreasing possible hampered discussed: may decrease release osteogenic factors osteoclasts, and/or released osteoclasts act directly neighboring cells as reported preclinical studies.

Bone is a highly specialized supporting framework of the body, characterized by its rigidity, hardness, and power of regeneration and repair. It protects the vital organs, provides an environment for marrow (both blood forming and fat storage), acts as a mineral reservoir for calcium homeostasis and a reservoir of growth factors and cytokines, and also takes part in acid–base balance (Taichman ...

Bisphosphonates are pyrophosphate analogues which were used for over a century in industry (mainly in the textile and oil industries) as antiscaling and anticorrosive agents because of their property of inhibition of calcium carbonate precipitation. After the discovery of biological effects of bisphosphonates more than 30 years ago, they have now become indispensable in medicine for the treatme...

Bisphosphonates are powerful drugs. Their ability to reduce the risk of osteoporotic fractures has saved thousands of people from becoming a fracture patient. Osteoporosis is due to an imbalance in the maintenance remodeling of the skeleton. This remodeling is different from the repair processes following fracture or bone surgery. Because research on the use of bisphosphonates has mostly focuse...

The vast majority of the longitudinal growth of a typical mammalian long bone occurs at the ends of the bone in platelike structures termed growth plates. Both growth plates, one at the proximal end and the other at the distal end, are peripheral extensions of the primary center of ossification, a structure that arises in the midportion of the cartilaginous anlage of the bone-to-be during early...
